Cayman's STEP Reporter Assays (Luminescence) consist of a 96-well plate pre-coated with a transfection complex consisting of the gene for the protein target of interest (i.e. an assay specific target protein), a SEAP reporter construct, and, in some cases, other genes needed to make the assay function with high fidelity. The plates are provide sealed and ready to use - all you need to do is add your cells. Transfection occurs overnight; cells can be tested the next day. The SEAP reporter is secreted into the media, so cellular responses can be followed over time. The kits are easily adapted to high throughput screening.

Additional info: The melanocortin-4 receptor (MC4R) has important roles in weight regulation, sexual function, and inflammation. Mice deficient in MC4R have increased lipid deposition associated with elevated adiposity, while mutations in MC4R in humans are associated with early onset or severe obesity. This assay consists of a 96-well plate coated with both MC4R and Secretory Alkaline Phosphatase (SEAP) reporter constructs. Cells grown on the STEP complex will express MC4R at the cell surface. Binding of agonists to MC4R initiates a signaling cascade resulting in expression of SEAP. SEAP activity is measured following addition of a luminescence-based alkaline phosphatase substrate provided in the kit.
