Flow Cytometry
Flow cytometry is used in a wide range of applications, including clinical testing and point-of-care diagnostics, genetics research, agricultural and veterinary sciences, food processing, and water purification testing. It is a fast, laser-based method for detecting pathogens or cancerous cells in a liquid, such as blood serum, and it can also be used to separate individual cells or molecules, for instance DNA amplified by PCR.
The technique of flow cytometry measures the number and type of particles that have been tagged or labeled with fluorescent dye molecules as part of a clinical assay or other lab procedure. A flow cytometer nozzle shoots a focused stream of liquid containing the assayed molecules at the rate of thousands of particles per second across a laser beam of a given wavelength. Each molecule in the liquid passes one-by-one through the beam, scattering the light. At the same time, the fluorescent tags on some of the molecules become energized by the light rays, giving off light at a longer wavelength than the laser beam.
The scattered and excited light rays are captured by electro-optical detectors, which convert the photons into electrical signals. These signals can then be analyzed by a computer to determine the composition of the molecules being tested.
Several different tags or dyes can be multiplexed in flow cytometery, to detect different molecules in the same operation. Some of the dyes can be “reversed,” or chemically removed from tagged molecules, in order to recycle them back into a patient’s blood stream, for example in cell therapy.
Laboratory equipment and supplies for flow cytometry include lasers and optical detectors; clinical and research laboratory assay kits; antibodies, proteins and peptides; antibody specific tags, conjugates (dyes) and dye combinations; staining or isolation reagents; and particle count standards.
