Nucleic Acid Delivery
Nucleic acids are large molecules with strong negative charges, making it difficult or impossible for them to navigate pores in cell and nuclear membranes. In order to introduce nucleic acids, such as RNA, DNA, proteins and antibodies into cells for clinical testing and research, as well as to safely and effectively apply genetic therapies in vivo, many strategies for transfection of nucleic acids to target locations have been developed.
Synthetic nucleic acid molecules, such as small interfering RNA (siRNA), can suppress the production of disease-related proteins, for example in tumors. To operate in vivo, siRNA needs protection from nucleases, as well as from the body’s other natural protection systems. Delivery methods must consider half-life in the blood-stream, tissue specificity, toxicity and efficiency. Some proposed methods include topical delivery of gold nanoparticle-nucleic acid conjugates through the skin, and encapsulating nucleic acid molecules within liposomes. These can travel safely through the bloodstream, to be taken into a tumor or infected cell by fusing with the cell membrane, and then depositing the molecules inside the cell.
Other nucleic acid delivery methods for clinical testing and research include magnetic-assisted transfection, viral transduction, plasmid DNA injection to mouse livers, electrotransfer, endocytocis (absorption) of cationic polymers, and calcium phosphate uptake.
Reagents and kits for nucleic acid delivery include a nanoparticle transfection reagent kit; a plasmid DNA transfection reagent; custom oligonucleotide synthesis; mycoplasma removal agent; cell culture media; growth containers; and DNA, RNA and protein extraction and purification kits.
Equipment for nucleic acid delivery includes real-time PCR systems; sample coolers; thermal cyclers; tissue homogenizers, centrifuges; hybridization incubators; microplate readers; and a nucleic acid purification instrument.
