a-Dystroglycan (Cranin)

Dystroglycans are essential elements of the neuromuscular junction (NMJ). The gene for dystroglycan is expressed as a precursor protein that is post translationally cleaved into a 156kD extracellular peripheral membrane protein called alpha dystroglycan and a 43kD transmembrane protein, beta dystroglycan. The latter protein contains a PPxY motif that promotes binding to WW domain containing proteins, such as utrophin and dystrophin. Phosphorylation at tyrosine 892 within the PPxY motif may regulate c-Src interactions with beta dystroglycan, as well as inhibit interactions with WW domain proteins. In skeletal muscle, beta dystroglycan is normally localized to the plasma membrane, however phosphorylation of Tyr892 leads to localization of beta dystroglycan to endosomal compartments along with c-Src. Thus, phosphorylation at Tyr892 may have important roles in altering the localization of beta dystroglycan during NMJ formation\n\nApplications: \nSuitable for use in Inhibition and Western Blot. Other applications not tested.\n\nRecommended Dilutions: \nInhibition of Laminin Binding to Dystroglycan: In a nitrocellulose overlay experiment, this antibody inhibits binding of 125I Laminin to Dystroglycan.\nWestern Blot: 1:1000-1:2000 Detects alpha-Dystroglycan in rabbit skeletal muscle. Rabbit skeletal muscle lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with D9905-05. Proteins were visualized using an IgM (HRP) goat anti-mouse and a chemiluminescence detection system.\nNote: The use of WGA puri?ed protein results in signi?cantly cleaner blots and immunoprecipitates. Post-translational modi?cation of Dystroglycan causes band broadening. \nOptimal dilutions to be determined by the researcher\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
