Mouse/rat AFP (alpha-fetoprotein) specific PCR primers for RT-PCR (reverse transcription followed by polymerase chain reaction) analysis of mRNA expression. \n\nComponents:\nPrimer Pair: Lyophilized. Each vial contains 375 pmoles of each primer. Adjust to a final concentration of 7.5 pmoles/ul by resuspending Primer Pair in either 50ul autoclaved deionized water or 0.1X TE buffer (1mM Tris HCl, pH8.0 @ 25 degrees C; 0.1mM EDTA, pH 8.0 @ 25 degrees C). \n\nPositive Control 59: Lyophilized. Each vial contains 150 ng synthetic double-stranded DNA. Resuspend in 30ul autoclaved deionized water or TE buffer. To prevent contaminating samples and reagents, it is strongly recommended that the Positive Control is resuspended in a separate location from where PCR reactions are set up. Use a different pipette than one used for PCR set up. The Positive Control is not intended for quantitative purposes.\n\nGenBank Accession Number: \nNM_007423 (mouse) and NM_012493 (rat) \n\nProduct Sizes: \ncDNA: 451 base pairs (bp) (mouse and rat) \nGenomic DNA: 3435 bp product predicted for mouse. Sequence not available for rat. No product observed for mouse or rat. \nPositive Control 59: 290 bp \nAlternate splice variants: None reported. \n\nSuggested Reaction Conditions: \nFor each 50ul Polymerase Chain Reaction mixture, 2ul of the primer pair stock solution should be used. Sufficient reagent is supplied for 25 separate amplification reactions of the gene fragment.\n\nThermocycling Conditions: \nA typical 50ul PCR would involve a 4 minute denaturation step at 94C, followed by 30-35 cycles of 45 seconds at 94C, 45 seconds at 55C and 45 seconds at 72C, with a final 10 minute extension step at 72C.\n\nStorage and Stability:\nThe Primer Pair resuspended in water or TE buffer is stable for up to one year at -20 degrees C in a non-frost free freezer. Aliquot in single use portions. Do not use past the expiration date above. Avoid repeated freeze-thaw cycles.
