D10 has been characterized in the ISOBM TD-2 workshop and assigned by K. Nustad to group D of a cluster of 6 major epitopes of human alpha fetoprotein. Its gene has been cloned and sequenced and its structure identified. AFP is expressed in fetal liver and is normally absent in healthy adult tissues. It is expressed in all yolk sac tumors, in some germ cell tumors and in hepatocellular carcinomas. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Frozen, formalin fixed paraffin sections Optimal dilutions to be determined by the researcher. Hybridoma: P3X63-Ag8.653 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
