The serum glycoprotein a1-antitrypsin (a-1 AT) is a typical member of the serpine family of serine protease inhibitors, and an inhibitor of the enzyme trypsin. a-1 AT is synthesized in the liver as a single polypeptide chain of approximately 51kD showing a marked degree of genetic polymorphism due to the presence of over 20 different alleles. Individuals homozygous for a-1 AT deficiency are predisposed to pulmonary emphysema and juvenile hepatic cirrhosis because of alterations in the amino acid and sialic acid components of the glycoprotein.\n\nApplications: \nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nOptimal dilutions to be determined by the researcher.\n\nCulture Medium:\nRPMI 1640 with 2-10% fatal calf serum\n\nHybridoma: \nX63-Ag8.653. Female CF1 x Balb/c mice immunized i.p. with immunogen adsorbed onto Al(OH)3.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes human a-1 AT, native and denatured. Epitope specificity differs from that of clone 6F4. Reacts strongly with a-1 AT. Strong reaction is seen in ELISA with native and denatured a-1 AT directly coated onto the microtiter well. When tested in sandwich ELISA in combination with a polyclonal antibody against a-1 AT, it only recognizes native a-1 AT. In Western Blot after SDS-PAGE, this antibody reacts with native and denatured a-1 AT in reduced as well as non-reduced forms.
