a2-Macroglobulin (alpha-2-Macroglobulin, Alpha-2-M, A2m, Pregnancy Zone Protein, Pzp)

Alpha-2 macroglobulin ids the major protein in the alpha-2 region of mouse serum after separation by electrophoresis. It is a large polymer and a strong antigen. It forms about one-third in the total of the alpha-2 globulins. The protein is isolated by ammonium sulphate precipitation, ion exchange chromatography, gel filtration and preparative starch block electrophoresis. In the purification of alpha-2 macroglobulin the activation of kallikrein is avoided, since it forms an irreversible bond with alpha-2 macroglobulin, thus contaminating the final immunogen.\n\nApplications:\nSuitable for use in IEP and RID. Other applications have not been tested. \n\nRecommended Dilutions: \nIEP: Use 2ul serum against 120ul antiserum\nRID: Use a rosette arrangement with 10ul antiserum in 3 mm diameter centre well and 2ul serum samples (neat and serially diluted) in 2 mm diameter peripheral wells. \nOptimal dilutions to be determined by the researcher. \n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes mouse alpha2 Macroglobulin. A single precipitin line is obtained with normal serum which shows a reaction of identity with the precipitin line with purified alpha-2 macroglobulin. Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other serum proteins. Does not crossreact with bovine, feline, chicken, canine,\nferret, goat, guinea pig, hamster, equine, human, monkey, rabbit, sheep, porcine or turkey.
