ATP-binding cassette (ABC) transporter genes are involved in the regulation of the amount of dietary cholesterol retained in the body. The ABC transporter ABCG1 (ABC8) is expressed on the cell surface and in intracellular compartments of cholesterol-laden macrophages and is induced in monocyte-derived macrophages during cholesterol influx mediated by acetylated low-density lipoprotein. In contrast, lipid efflux in cholesterol-laden macrophages, mediated by the cholesterol acceptor HDL(3), suppresses the expression of ABCG1.\n\nApplications:\nSuitable for use in Western Blot and Immunohistochemistry. Not recommended for Immunoprecipitation.. Other applications not tested. \n\nRecommended Dilution: \nWestern Blot: Mouse peritoneal macrophages treated with 22-R-hydroxycholesterol and T0901317 (stimulating ABCG1 expression). Recognizes a band at ~75.6 kD\nOptimal dilution determined by the researcher. \n\nStorage and Stability:\nStore at 2-8 degrees C. Do Not Freeze! Aliquots are stable for at least 3 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
