Alpha-actinin is a ubiquitously expressed protein found in numerous actin structures. It consists of an N-terminal actin binding domain, a central rod domain, and a C-terminal domain and functions as a homodimer to cross-link actin filaments. The rod domain determines the distance between cross-linked actin filaments and also serves as an interaction site for several cytoskeletal and signaling proteins (1). Alpha-actinins are a multigene family of four actin-binding proteins related to dystrophin. Alpha-actinin-1 and -4 (ACTN1, ACTN4) are non-muscle isoforms version of ACTN. These isoform are commonly expressed in filopodia, lamellipodia, stress fibers, sites of adhesion and intercellular contacts (2). Applications: Suitable for use in Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:2,000-10,000 Immunohistochemistry: 1:100-250 Flow Cytometry: 1:80 Immunoprecipitation: 1:40 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Manufactured incorporating RabMAb(R) technology under Epitomics US patents, No 5,675,063 and 7,429,487, owned by Abcam.
