Alpha-actinin is a ubiquitously expressed protein found in numerous actin structures. It consists of an N-terminal actin binding domain, a central rod domain, and a C-terminal domain and functions as a homodimer to cross- link actin filaments. The rod domain determines the distance
between cross-linked actin filaments and also serves as an interaction site for several cytoskeletal and signaling proteins (1).Alpha-actinins are a multigene family of four actin- binding proteins related to dystrophin. Alpha-actinin-1 and -4 (ACTN1, ACTN4) are non-muscle isoforms version of ACTN. These isoform are commonly expressed in filopodia, lamellipodia, stress fibers, sites of adhesion and intercellular contacts (2).
Applications:
Suitable for use in Flow Cytometry, ELISA, Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested.
Recommended Dilution:
Flow Cytometry: 1:40
Western Blot: 1:500-1:1000
Immunoprecipitation: 1:40
Immunocytochemistry: 1:100-1:250
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
