The activating transcription factor ATF2 (also called CRE-BP1) binds to both AP-1 and CRE DNA response elements and is a member of the ATF/CREB family of leucine zipper proteins. ATF2 has been implicated in the transcriptional regulation of a number of genes including cytokines, cell cycle control and apoptosis. Various forms of cellular stress, including inflammatory cytokines and UV irradiation, stimulate the transcriptional activity of ATF2. Stress induced ATF-dependent transcription is dependent on phosphorylation of ATF. Serine 490 and serine 498 are novel phosphorylation sites on ATF that have recently been identified. In addition, ATF2 is particularly abundant in the brain and the ATF2 family of transcription factors is considered an important substrate of signals upstream of the activation of genes associated with neuronal growth and differentiation. ATF expression has also been linked to the depression in humans. Applications: Suitable for use in Dot Blot, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Dot Blot: 1:1000 Western Blot: 1:1000 Immunohistochemistry: 1:1000 Note: Antibody dilutions and tissue load should be based on tissue type and expected phosphorylation state expected expression level. Optimal dilutions to be determined by the researcher. Cellular Localization: Nuclear Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
