Activating Transcription Factor 6 (ATF6, ATF6-alpha, ATF6A, Cyclic AMP-dependent Transcription Factor ATF-6 alpha)

ATF6, a basic-leucine zipper protein, was isolated by yeast interaction assay as a binding partner to SRF (serum response factor). SRF is a transcription factor, which binds to the serum response element (SRE) in the c-fos promoter and regulates expression of the c-fos gene as well as other immediate-early genes and some tissue-specific genes. ATF6 has also been shown to interact with the endoplasmic reticulum (ER) stress response element (ERSE). ATF6 constitutively expressed as a membrane-bound 90kD protein is directly converted to a 50kD protein in ER-stressed cells. When unfolded proteins accumulate in the ER, ATF6 is released and translocates to the golgi where it is cleaved into its active form which enters the nucleus and activates translation of UPR genes including XBP1.\n\nApplications:\nSuitable for use in Western Blot, Immunohistochemistry, Immunocytochemistry and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1-5ug/ml. Detects bands of ~90kD (full length ATF6) and ~50-70 kD (cleaved form of ATF6).\nImmunohistochemistry: Paraffin and frozen sections\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
