ADAM12 is a 120kD type I transmembrane glycoprotein member of the metzincin family of proteases. It is synthesized as a 909aa preproprecursor that contains a 25kD proregion aa29-207 and a 92kD mature molecule. Proteolytic cleavage in the Golgi removes the propeptide prior to cell-surface expression. A 68kD soluble splice form exists in pregnancy where 36aa replace the C-terminal 205 aa705-909. This circulating ADAM12 isoform remains associated with its prodomain via noncovalent linkage, possibly acting as an enzyme regulator. Over aa29-513, human ADAM12 shares 78-79aa identity with bovine and mouse ADAM12, respectively.
Applications:
Suitable for use in Immunoprecipitation. Other applications not tested.
Recommended Dilution:
Immunoprecipitation: 25ug/ml detects ADAM12 in conditioned cell culture medium spiked with recombinant human ADAM12.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
Lyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
