Human SH2 domain containing leukocyte protein-76-Associated Phosphoprotein of 130kD (SLAP-130), also known as Fyb-120 and ADAP, is an 86kD, 783 amino acid, cytoplasmic protein that runs anomalously as a 120-130kD protein in SDS-PAGE. Human SLAP-130 is a hematopoietic-specific adaptor that associates with other cytoplasmic proteins such as SH2-containing Leukocyte Phosphoprotein of 76kD (SLP-76). Adaptors integrate signals from surface receptors and are essential components of many immune cell-signaling cascades. There is at least one known alternatively-spliced SLAP-130 isoform which contains an additional 46 aa. Over the region used as immunogen, SLAP-130 is 66% and 76% identical to the corresponding canine and mouse proteins, respectively.
Applications:
Suitable for use in Flow Cytometry and Direct ELISA. Other applications not tested.
Recommended Dilution:
Flow Cytometry: 25ug/mL use to 10ul to label 10e6 cells
Direct ELISA: 0.5-1ug/mL detection limit for rhSLAP-130 ~ 5ng/well.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
Lyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
