FYN-binding protein (FYB) is an adapter protein for Fyn and SLP-76 signaling cascades in T cells. A hematopoietic protein, FYB associates with p59fyn and becomes tyrosine-phosphorylated in response to receptor ligation. FYB has been associated to modulation of IL-2 expression in T cells and platelet activation (3). Fyn (p59Fyn) is a 59kDa member of the Src family of tyrosine kinases. The carboxy terminus of Fyn shares extensive amino acid sequence homology with Src, but is very different within the amino-terminal 81 amino acid residues. The Fyn protein is synthesized and N-myristoylated on cytosolic polysomes and then rapidly targeted to the plasma membrane, where it is palmitoylated. Fyn's biological functions are diverse and include signaling via the T cell receptor, regulation of brain function and adhesion mediated signaling (2, 3). Suitable for use in Flow Cytometry, Western Blot, Immunohistochemistry, Immunocytochemistry. Other applications not tested.
Recommended Dilution:
Western Blot: 1:5000-10,000
Immunohistochemistry: 1:250-500
Immunocytochemistry: 1:100-250
Flow Cytometry: 1:60
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
