Adenovirus, Type 5, Early Region (E1A) (BSA & Azide Free)

The early region (E1A) of the adenovirus genome plays a central role in cellular transformation and regulation of gene expression. The adenovirus early gene product E1A is a potent stimulator of cellular proliferation, which when overexpressed can overcome the growth-inhibitory effects of TGF-beta. The E1A region encodes a series of related proteins (35-46kDa) with multifuctional capabilities and form a specific complex with the retinoblastoma tumor suppressor gene product (Rb protein). The E1A and E1B regions together comprise the transforming region of adenovirus. While expression of E1A alone is sufficient to immortalize primary cells, complete transformation requires the additional expression of the E1B region. Several conserved regions of E1A are similar to portions of other viral oncoproteins like the HPV-16 and HPV-18 E7 and SV40 large T antigen. \n\nApplications: \nSuitable for use in Immunofluorescence, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilution: \nImmunofluorescence: 0.2-0.4ug/ml\nImmunoprecipitation (Use Protein A): 2ug antibody/mg protein lysate\nWestern Blot: 0.5-1.0ug/ml\nImmunohistochemistry: 2-4ug/ml for 30 min at RT.\nNote: Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\n293 cells, adenovirus infected cells and tissues.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C or colder. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
