Adenovirus 5 belongs to Adenoviridae, a family of viruses with at least 51 serotypes that cause a variety of illnesses, mostly being upper respiratory tract infections. Adenovirus type 5 antigen is collected from both the cell and supernatant phases of tissue culture. The antigen is harvested by disrupting optimally infected monolayers in tissue culture medium. Particles are purified by double caesium chloride centifugation, providing an ultrapure preparation. The antigen preparation is inactivated by heat treatment at 54 degrees C for 2 hours. Inactivity is confirmed by growth on 293 cells for two weeks with no CPE seen.
