Agouti is a paracrine-signaling molecule that affects pigmentation by inhibiting the melanocortin receptor 1 (MCR-1). However, recombinant Agouti protein also antagonizes the MC2R and MC4R. AGRP (132 aa in human, chromosome 16q21) is normally expressed in adrenal and hypothalamus. AGRP levels are increased several folds in ob/ob mice. AGRP is a strong antagonist of MC3R and MC4R. Ubiquitous expression of AGRP in transgenic mice causes obesity without altering skin pigmentation. Applications: Suitable for use in Immunohistochemistry and Immunofluorescence. Other applications not tested. Recommended Dilution: Immunofluorescence: 1:200 Immunohistochemistry (PAP or ABC): 1:500 Optimal dilutions to be determined by the researcher. AA Sequence: Ser-Pro-Arg-Arg-Cys-Val-Arg-Leu-His-Glu-Ser-Cys-Leu-Gly-Gln- Gln-Val-Pro-Cys-Cys-Asp-Pro-Cys-Ala-Thr-Cys-Tyr-Cys-Arg-Phe- Phe-Asn-Ala-Phe-Cys-Tyr-Cys-Arg-Lys-Leu-Gly-Thr-Ala-Thr-Asn- Leu-Cys-Ser-Arg-Thr-NH2 (Five S - S bonds formed, disulfide bridges undetermined) Storage and Stability: Lyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
