Produced in goats immunized with purified, insect cell line Sf 21-derived, recombinant human Agouti-Related Protein (rhAgRP), also known as the protein product of Agouti-related transcript (ART). AgRP specific IgG was purified by human AgRP affinity chromatography and then biotinylated.
Applications:
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilution:
ELISA: Can be used as a detection antibody in a human AgRP ELISA in combination with the monoclonal capture antibody (Catalog# A1059-62G). Using plates coated with 100ul/well of the capture antibody, in combination with 100ul/well of the detection antibody at 50ng/ml, an ELISA for sample volumes of 100ul can be obtained. To arrive at the optimal dose range for
this ELISA, set up a two-fold dilution series of the protein standard stAgRPing with 4ng/ml.
Western Blot: 0.1-0.2ug/ml. The detection limit for rhAgRP is approximately 1ng/lane under non-reducing and reducing conditions.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
Lyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
