Phosphorylation site specific antibody selective for the phosphorylated form of the AKT protein containing a phosphate on threonine 308. The antibody has been shown to recognize AKT in NIH-3T3 cells stimulated with PDGF. AKT is a key effector of the PI3K pathway. Several studies have shown that a signalling pathway from PI3K to AKT may mediate some cellular responses of PI3K, including protection from apoptosis in PC12 cells. AKT accomplishes its cell survival-promoting function by inhibiting apoptosis by means of its ability to phosphorylate and inactivate BAD and caspase 9. The function of AKT is regulated by phosphorylation on two sites conserved within the AGC kinase family: the activation loop (threonine 308) in the kinase core and hydrophobic phosphorylation site on the carboxyl terminus (serine 473). Threonine 308 is phosphorylated by the phosphoinositide-dependent kinase 1 (PDK 1). Cellular Localization: Cytoplasmic and Nuclear Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
