Akt Substrate, 160kD, phosphorylated (Ser666) (AS160, DKFZp779C0666, KIAA0603, TBC1 domain family member 4, Rab GAP)

AS160 (Akt Substrate of 160kD, TBC1D4) is a Rab-GAP protein that regulates the trafficking of GLUT4 (glucose transporter 4) to the plasma membrane. AS160 is known to be phosphorylated on at least 8 residues following stimulation from IGF-1, EGF, PMA, and AICAR (Chen, S., 2008). Thr642 is phosphorylated by Akt in response to IGF-1, while Ser341 and Thr642 of AS160 are phosphorylated by RSK (p90 ribosomal S6 kinase) in response to PMA. Ser588 has been shown to be phosphorylated by AMPK in vitro, though other kinases might be responsible as well. The phosphorylation site, Ser666, does not contain the Akt consensus sequence. Its phosphorylation is suppressed by AICAR and by combined mutations of TOS (mTOR signaling)-like sequence (FEMDI) and rapamycin (Geraghty, K.M., 2007). Additionally, its phosphorylation is markedly increased in response to IGF-1 and this phosphorylation is blocked in response to the PI3 Kinase inhibitor LY294002 (Geraghty, K.M., 2007).\n\nApplications:\nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:250\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
