Akt1, PH Domain (Rac PKa, PKBa)

The serine/threonine specific protein kinase, Akt1/PKBa/RAC-a is the cellular homolog of the viral oncogene v-akt. Growth factor or insulin stimulation of mouse NIH 3T3 fibroblasts or Rat-1 cells activates Akt1. Activation of Akt1 is concomitant with phosphorylation of Thr308 and Ser473 and is prevented by inhibitors of phosphatidylinositol 3-kinase. PtdIns (3,4,5)P3-dependent kinase-1 (PDK-1) phosphorylates Akt1 on Thr308. Akt1 has been shown to inhibit insulin-induced stimulation of glycogen synthase kinase-3 (GSK-3) activity. \n\nApplications:\nSuitable for use in Western Blot.. Other applications not tested.\n\nRecommended Dilution:\nImmunoblot Analysis: 1:500-1:2000 dilution detects Akt1/PKBa in RIPA lysates from mouse 3T3 cells. Mouse NIH 3T3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Akt1/ PKBa, PH Domain \n(1:1000). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.\n\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
