Amphiphysin I is a 76kD protein with an apparent molecular mass of 128kD, that associates with the cytoplasmic surface of synaptic vesicle (1). Amphiphysin is highly enriched in nerve terminal, where it has a putative role in clathrin-dependent recycling of secretory vesicle (2). Amphiphysin binds dynamin 1 and synaptojanin 1 through carboxy terminal SH3 domains (3). Additional interactions have been observed with clathrin and AP-2. It is through these interactions that amphiphysin is thought to play a role in clathrin mediated endocytosis. Amphiphysin is the dominant autoantigen in paraneoplastic Stiff-Man syndrome (4).\n\nApplications:\nWestern Blot (Colorimetric): 0.5ug/ml\nImmunoprecipitation\nImmunohistochemistry: 5ug/ml\nOptimal dilutions to be determined by the researcher.\n\nPositive Controls: \nRat Brain Tissue Extract\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
