AMPK beta1, phosphorylated (Ser108) (AMP-activated Protein Kinase)

AMP-activated protein kinase (AMPK) is highly conserved from yeast to plants and animals, and plays a key role in regulation of energy homeostasis (1). The kinase is activated by an elevated AMP-ATP ratio due to cellular and environmental stress, such as heat shock, hypoxia and ischemia (2). AMPK also has a role in apoptosis (3). AMP plays an essential role in the activation of AMPK. Firstly, it is required for AMPK phosphorylation by the upstream kinase AMPKK at Thr172 (4). AMPK phosphorylation by AMPKK raises AMPK\nactivity at least 50-fold. Secondly, AMP also inhibits the dephosphorylation and inactivation of AMPK by PP2C (5). The beta1 subunit is post-translationally modified by myristoylation and multi-site phosphorylation. Phosphorylation at serine 108 of the beta1 subunit has\nbeen suggested to enhance the activity of AMPK enzyme (6). Phosphorylation at serines 24, 25 and 182 of the beta1 subunit affects its cellular localization (6).\n\nApplications: \nSuitable for use in Western Blotting, Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:1000\nImmunoprecipitation: 1:50\n\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (30-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer
