Amphiregulin (AR) binds to EGF-Receptor (EGFR) with lower affinity than EGF. The mature secreted form AR is an 84aa residue glycosylated polypeptide growth regulator, which is generated by proteolytic processing of a 252aa transmembrane precursor. Seven different polypeptide ligands, which derive from distinct genes, are capable of binding to the extracellular domain of EGFR. These ligands include EGF, TGFa, AR, HB-EGF, cripto-1, epiregulin, and betacellulin. All of these growth factors contain a characteristic EGF-like domain, which is defined by 6 evenly spaced cysteine residues that generate 3 loops through the formation of disulfide bonds. AR protein has been localized by immunohistochemistry to the epithelium of the colon, stomach, pancreas, breast, and placenta. AR is reportedly overexpressed in human cancers of the breast, colon, stomach, and pancreas.

Recommended Dilution:
Western Blot: 2.5-5g/ml
Immunohistochemistry: 1:200 for 10min at RT
Staining of formalin-fixed tissues requires boiling tissues sections in 10mM citrate buffer, pH 6.0, for 10min followed by cooling at RT for 20min

Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
