Amyloid Precursor Protein, CT (APP, Amyloid beta (A4) Precursor Protein)

The APP is a large transmembrane protein which is constitutively expressed by many cell types. Within the Golgi apparatus and endosome, APP is oriented such that its N-terminus is within the lumen while the C-terminus extends into the cytoplasm. Cleavage of APP by b-secretase (BACE) generates the N-terminus of the b amyloid peptide within C99, the C-terminal fragment. Further cleavage by g-secretase yields the b amyloid peptide. b amyloid is released into the extracellular space. \n\nApplications: \nSuitable for use in ELISA, Western Blot. Other applications not tested.\n\nRecommended Dilution:\nELISA: (same/more than)1:25,000\nWestern Blot: (same/more than)1:5,000\nOptimal dilutions to be determined by the researcher.\n\nRecommended Positive Control: Neuronal cell lysate.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the ori0ginal vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes the C-terminus of the human Amyloid Precursor Protein (APP), a protein with Mr 100kD. The smaller APP C-terminal fragment b (CTFb; ~8kD) is rapidly degraded and not detected by Western blot analysis of brain cell lysates. May crossreact with Amyloid Precursor-Like Protein2 (APLP2). This antiserum can routinely be used without further purification.
