Angiotensin Converting Enzyme (ACE, CD143)

Human CD143, also known as angiotensin-converting enzyme (ACE). The somatic form is expressed by endothelial cells (especially those of arterioles and lung capillaries), epithelial cells (especially in proximal renal tubules and in the small intestine), by some neuronal cells and variably on some macrophages and T lymphocytes. The germinal form is expressed by spermatozoa. Study of CD143 may be relevant to fertility (especially sperm-egg interactions), atherosclerosis and in drug targeting to the lung. This clone has been reported to recognize ACE in Western Blot. \n\nApplications:\nSuitable for use in Immunohistochemistry, Western Blot and \nELISA. Other applications not tested.\n\nRecommended Dilutions:\nImmunohistochemistry (Paraffin): 1:10-1:100. Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended.\nWestern Blot: 1:50-1:100\nOptimal working dilutions to be determined by researcher.\n\nRecommended Positive Control Tissue: \nHuman Kidney \n\nRecommended Negative Controls: \nI1904-79G: Mouse IgG1 Negative Control\n\nRecommended Secondary Reagents: \nF(ab')2 rabbit anti-mouse IgG HRP \n\nHybridoma: \nSpleen cells from immunized Balb/c mice were fused with cells of the mouse X36-Ag8-653 myeloma cell line.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol 40-50%, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes human CD143, also known as angiotensin-converting enzyme (ACE). CD143 exists in two forms, a 170kD somatic form and a 90kD germinal form. The antibody recognizes denatured ACE, binding to an epitope in the C domain. \nSpecies Crossreactivity: monkey, rabbit, bovine, canine, feline, guinea pig, goat and sheep. Does not react with porcine and rat.
