Angiotensin Converting Enzyme, Bovine (ACE)

ACE (kininase II, EC 3.4.15.1) is a zinc dipeptidyl carboxypeptidase (1). ACE catalyses both angiotensin I transformation to angiotensis II (a potent vasopressor agent) and degradation of bradikinin, a vasodepressor. Thus, ACE appears to play a key role in regulating vascular tone and remodeling as well as in the development of atherosclerotic lesions. Increasing evidence asuggests that ACE plays an important role in vascular pathology. Recent studies show two new functions for this important enzyme: 1) ACE enhances presentation of endogenous antigens (such as HIV gp160-derived peptide p18) to MHC class I-restricted T-lymphocytes (1); 2) amino-terminal domain of ACE efficiently cleaves hemoregulatory tetrapeptide N-Ac-Ser-Asp-Lys-Pro (2,3) which is involved in the control of haemopoetic stem cell proliferation (the amount of this peptide greatly decreases in patients undergoing cancer chemotherapy) (4).\n\nSpecific Activity: ~15mU/ug of protein. \n\nUnit Definition: One unit produces 1umole of hippuric acid or His-Leu from Hippuril-His-Leu per min. in 0.1M phosphate buffer, 300mM sodium chloride, pH 8.3 at 37 degrees C.\n\nInhibitors: Metalloproteinase inhibitors such as EDTA and 1,10-phenanthroline.\n\nStable pH: 7.5-8.5.\n\nApplications: Suitable for use as a reference in SDS-PAGE, Western Blot [See A2295-01B and A2295-01G. Use 20ng/lane] and Immunoprecipitation. Also suitable for enzymatic studies including substrate specificity and enzyme kinetics. Other applications not tested.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
