Control Peptide for A2295-07D (antiserum) and A2295-07E (affinity purified antibody). Angiotensin II interacts with two types of G-protein coupled membrane receptors, AT1 (type 1) and AT2 (type 2). Major cardiovascular effects appear to be mediated through AT1. AT1 has three isoforms (rat AT1A 359aa; AT1B/AT III, 359aa; and AT1C, 177aa). Rat AT1 receptors are predicted to contain seven transmembrane domains. The N-terminus is predicted to be extracellular. The C-terminus is inside the membrane (cytoplasmic). The binding of angiotensin II with AT1 receptors activate a phosphatidylinositol-calcium second messenger system. AT1 receptors are expressed in the liver, kidney, aorta, lung, uterus, ovary, spleen, heart, adrenal and vascular smooth muscle. AT2 gene ( chromosome x) encodes 363 aa protein. It is highly expressed in myometrium with lower levels in adrenal and fallopian tube. It is also expressed at high levels in fetal kidney and intestine. Rat AT1 receptors are predicted to contain seven transmembrane domains. The N-terminus is predicted to be extracellular, while C-terminus inside the membrane (cytoplasmic). Applications: Suitable for use in ELISA, Antibody Blocking, Not suitable for use in Western Blot due to low moleculare weight. Other applications not tested. Recommended Dilution: Antibody Blocking: 5-10ug per 1ul A2295-07D (antiserum) or 1ug A2295-07E (affinity purified antibody). ELISA: 50-100ng of control peptide/well. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional info: Synthetic peptide 10aa sequencemapping near the N-terminus of rat AT II (1). This peptide is predicted to be extracellular. Sequence is the same in all AT II receptor isoforms (Type 1a, 1b and 1c). No sequence homology with other G-protein coupled receptors.Species sequence homology: Mouse, rat, human, porcine and rabbit: 100%.
