Sacsin (UniProt Q9NZJ4; also known as DnaJ homolog subfamily C member 29, DNAJC29, Spastic ataxia of Charlevoix-Saguenay) is encoded by the SACS (also known as SPAX6; ARSACS, KIAA0730, PPP1R138) gene (Gene ID 26278) in human. Sacsin is a multimodular protein composed of an ubiquitin-like domain that binds to the proteosome, three large sacsin repeat regions that may have an Hsp90-like chaperone function, an XPCB domain that binds to the Ube3A ubiquitin protein ligase, a DnaJ domain that binds Hsc70, and a higher eukaryotes and prokaryotes nucleotide-binding domain that mediates sacsin dimerization. Sacsin is localized to mitochondria and plays an important role in mitochondrial morphology and function. Sacsin disruption leads to impaired organization of dendritic arbors and neuronal cell death. Autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS) is a childhood-onset neurological disease resulting from mutations in the SACS gene encoding sacsin.
