Suitable for use in Western blot, ELISA, immunofluorescence microscopy and immunocyto/histochemistry (frozen and paraffin-embedded). With paraffin-embedded sections, protease pretreatment is required prior to antibody application. Dilute at least 1:10 with PBS, pH 7.4 for immunohistochemical applications. Each laboratory should determine an optimum working titer for use in its particular application. IF, along withtubulin-based microtubules and actin-based microfilaments, comprise thecytoskeleton. All IF proteins are expressed in a highlydevelopmentally-regulated fashion; vimentin is the major cytoskeletalcomponent of mesenchymal cells. Because of this, vimentin is often used as amarker of mesenchymally-derived cells or cells undergoing an epithelial-to-mesenchymaltransition (EMT) during both normal development and metastatic progression.
