Antithrombin III (ATIII)

Antithrombin III (ATIII) is a single chain glycoprotein of molecular mass 58kD with four glycosylation sites and three intra-molecular disulfide bonds. AT III is normally present in human plasma at a concentration of approx. 12.5mg/dl. It is the major inhibitor of thrombin. Inactivation of thrombin by AT-III occurs by formation of a covalent bond resulting in an inactive 1:1 stoichiometric complex between the two components.\n\nApplications: \nSuitable for use in ELISA and Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nELISA: 1:32,000\nOptimal dilutions to be determined by the researcher.\n\nHybridoma: \nX63-Ag8.653. Female CF1 x Balb/c mice immunized i.p. with immunogen adsorbed onto Al(OH)3.\n\nCulture Medium:\nRPMI 1640 with 10% fetal calf serum.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes human ATIII, native form. Epitope specificity differs from those of clones 6F27 and 6F218. Strong reaction is seen in ELISA with native ATIII coated directly onto the microtiter well, and also when used as detection antibody in sandwich ELISA in combination with a polyclonal capture antibody. Reacts with ATIII in Western Blot after SDS-PAGE.
