Antithrombin III (ATH III) is a plasma protein synthesized in the liver. Normal plasma levels are 115 to 160mg/L. ATH III consists of a single polypeptide chain and has a molecular weight of 58kDa. ATH III is a serine protease inhibitor and regulates coagulation by the formation of covalently liked complexes. ATH III also has a heparin binding site and in the presence of heparin the inhibition of thrombin, factors IXa, Xa and XIa is enhanced 1000 fold. Low levels of ATH III (functional or concentration) are associated with deep vein thrombosis and pulmonary embolism. Antithrombin assays are based on immunologic, enzymatic, or clotting properties Applications: Suitable for use in RID, Double Difffusion and Rocket IEP. Other applications not tested. Recommended Dilutions: RID: Neat-1:10; 6ul antiserum/cm2 in gel vs 5ul fresh plasma Double Diffusion: 10ul antiserum vs 5ul fresh plasma IEP: 100ul antiserum vs 5ul fresh plasma Note: Use 3% PEG 6000 with 1.2% agarose in a suitable buffer (TBE or Tris-barbital pH > 8.2). Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Additional info: Recognizes human antithrombin III. A single arc is detected when tested by IEP and 2D-IEP against human plasma. Identity is confirmed by double diffusion (Ouchterlony) vs human plasma and a known anti-human antithrombin III. Species Crossreactivity: rat, equine, mouse, canine and feline. Does not react with rabbit, bovine, porcine, guinea pig or chicken.
