AP-2 alpha

AP-2 alpha is a 52kD retinoic acid-inducible and developmentally regulated activator of transcription that binds to the consensus DNA-binding sequence CCCCAGGC in the SV40 and metallothionein promoters. There are three different but highly homologous AP-2 isoforms: alpha, beta, and gamma, encoded by three different retinoic acid-responsive genes. The AP-2 proteins are normally expressed in ectodermally derived tissues where they are necessary for normal growth and development. The factors have also been implicated in the control of cell proliferation, viral transformation, and oncogenesis. AP-2 seems to play in important role in human breast cancer; all three isoforms of AP-2 have been shown to activate a c-erbB-2 report construct, though the alpha and gamma forms are reported to be 3-4 times more effective than the beta form. AP-2 alpha may also be a factor in predicting survival in patients suffering from epithelial ovarian cancer. AP-2 alpha is the only AP-2 protein required for early morphogenesis of the lens vesicle.\n\nApplications:\nSuitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nELISA: 0.1-1ug/ml\nImmunohistochemistry (paraffin): 0.5-5ug/ml\nWestern Blot: 0.5-1ug/ml\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Specific for the AP-2 alpha protein and does not react with AP2 beta or gamma. On Western blots, it identifies the target band at ~52kD. Based on amino acid sequence homology, reactivity with mouse and rat is expected. Reactivity has been confirmed with human HeLa cell lysates by Western blotting and with paraffin-embedded breast tumor and tumor-associated normal tissue by immunohistochemistry.
