The apelin receptor (APJ receptor) has been identified as the endogenous receptor for the the peptide apelin. Previous to the identification of apelin, APJ was considered an orphan G protein coupled receptor. The apelin receptor (both by immunohistochemistry and in situ) has been localized to the CNS and is currently thought to play a role in stress response, feeding behavior and haemodynamic homeostasis. However, because of a broad distribution the apelin receptor and its endogenous ligand be involved in a wide range of biological process yet to be determined.\n\nApplications: \nSuitable for use in ELISA and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nImmunohistochemistry: 1:10,000-1:50,000\nTransfected COS-7 cells, plated on ploy-L-lysine coated glass coverslips, were fixed for 20 min with 4% paraformaldehyde in 0.1M PB, pH 7.4, rinsed with 0.1M TBS, pH 7.4, and preincubated for 30 min at RT with a blocking solution consisting of 5% normal goat serum, 2%BSA, AND 0.1% Triton X-100 in 0.1M TBS. Cells were incubated overnight at 4 degrees C with anti-APJ peptide antiserum diluted to 1:10,000-1:50,000 in 0.1M TBS, pH 7.4 containing 1%NGS and 0.05% Triton X-100. After washing with TBS, cells were incubated for 1 hr at RT with appropriate secondary antibody, washed twice with TBS and mounted on glass slides with Aquamount.\nOptimal dilutions to be determined by the researcher.\n\nControl Peptides: \nA2298-80A, A2298-80B\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
