MEKK5, also known as ASK1 for apoptosis signal-regulating kinase-1, is a MAP3K type protein kinase and a member of the JNK and p38 MAP kinase signaling pathways. The protein contains a centrally located protein kinase domain, a N-terminal extension that includes binding regions for the Fas-associated adapter protein Daxx and the redox-sensing enzyme thioredoxin, and a C-terminal extension that binds members of the TNFR-associated factor (TRAF) family. MEKK5 regulates the JNK cascade by phosphorylating MKK4/MAP2K4 and p38 cascades by phosphorylating MEK3/MAP2K3 and MKK6/MAP2K6. MEKK5 has been shown to be activated by oxidant stress, tumor necrosis factor (TNF) via TRAF2 and Fas via Daxx. MEKK5 also can be activated by Fas-Daxx in a kinase-independent apoptotic pathway.
Applications:
Suitable for use in ELISA, Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested.
Recommended Dilution:
Immunohistochemistry (Formalin fixed paraffin embedded): 5ug/ml
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
