AG4B is a 393aa cysteine protease consisting of the active catalytic triad site of Cys74, His280 and Asp278 and belonging to the peptidase C54 family. It is required for autophagy, which is essential for cell homeostasis and cell remodeling during differentiation, metamorphosis, non-apoptotic cell death and aging. It negatively regulates the localization of LC3 to a membrane compartment by delipidation and mediates murine Atg8L/Apg8L modification during the formation of autophagosomes. APG4B is expressed in skeletal muscle, brain, heart, liver and pancreas.
