The ability of cells to undergo bulk degradation of endogenous proteins through an autophagosomic-lysosomal pathway, termed autophagy or Type II cell death, is important in the control of cell growth and development. Studies have identified Beclin-1 as a Bcl-2 interacting mammalian gene which not only mediates autophagy, but also functions as a tumor suppressor, being expressed at reduced levels in human breast carcinoma, and may also play a role in defense against viral infection. An increasing number of studies have revealed an interaction between proteins which induce autophagy and those involved in apoptosis and in some instances apoptosis may be dependent upon or delayed by autophagy. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.5ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
