KCa 1.1 (BK Ca , Maxi K +or slo) is part of a structurally diverse group of K +channels that are activated by an increase in intracellular Ca 2+ . K Ca 1.1 shows a large single channel conductance when recorded electrophysiologically and hence its name. It differs from the rest of the subfamily members in that it can be activated by both an increase in intracellular Ca 2+ and by membrane depolarization. KCa 1.1 is expressed in virtually all cell types where it causes hyperpolarization and helps to connect between intracellular Ca 2+ signaling pathways and membrane excitability. Indeed, K Ca 1.1 channels have a crucial role in smooth muscle contractility, neuronal spike shaping and neurotransmitter release.
Applications:
Suitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications have not been tested.
Recommended Dilutions:
Western Blot: 1:500. Rat brain membrane
Immunoprecipitation: 4ug. Rat brain lysate
Immunohistochemistry: Rat brain sections
Optimal dilutions to be determined by the researcher.
Control Peptide:
B2103-05A: Supplied as a lyophilized powder. The stock solution of the antigen can be made up using 100ul of sterile ddH20. For negative control, preincubate 1ug peptide with1ug of antibody for one hour at RT.
Storage and Stability:
Lyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
