BRAF, a member of the RAF subfamily of Ser/Thr protein kinases, is involved in the transduction of mitogenic signals from the cell membrane to the nucleus. It may play a role in the postsynaptic responses of hippocampal neurons. This cytoplasmic protein is expressed in brain and testis. Defects in BRAF are involved in a wide range of cancers including lung cancer and non-Hodgkin lymphoma (NHL). This protein contains 1 zinc-dependent phorbol-ester and DAG binding domain.
Applications:
Suitable for use in Immunofluorescence, ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions:
Immunofluorescence: 1:200-1:1000
ELISA: 1:10,000
Western Blot: 1:500-1:2000
Immunohistochemistry (FFPE): 1:200-1:300. Validated for use on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with B2680-01Y, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
