BDNF is a member of the nerve growth factor family of trophic factors. In the brain BDNF has a trophic action on retinal, cholinergic, and dopaminergic neurons, and in the peripheral nervous system it acts on both motor and sensory neurons. Some protein domains of BDNF are identical with those of NGF and another neurotrophic factor, designated NT-3 (neurotrophin-3 ). Polyclonal antibodies raised against murine NGF have been shown to crossreact with both NT-3 and BDNF. Recombinant Human Brain-Derived Neurotrophic Factor is a homodimer, non-glycosylated, polypeptide chain containing 2 x 119 amino acids and having a total molecular mass of 26.98kD. Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-His-Ser-Asp-Pro. Biological Activity: Recombinant BDNF is fully biologically active when compared to standard. The ED50, calculated by the dose-dependent induction of ACHE (acetylcholine esterase) in rat basal forebrain primary septal culture is 50ng/ml. Protein Content: Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280nm using the absorbency value of 1.6 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a standard solution of BDNF as a Reference Standard. Storage and Stability: Lyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute by adding sterile ddH2O. For long term it is recommended to add a carrier protein (0.1% HSA or BSA). Aliquot and store at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
