In cardiac tissue brain natriuretic peptide (BNP) is synthesized as 134 amino acid precursor (prepro-BNP), which is cleaved by proteases to form a 26 aa "signal" peptide and a 108 aa pro-BNP. Proteolytic digestion of pro-BNP results in formation of 76 aa amino-terminal NT-proBNP and biologically active 32 aa BNP hormone molecule. Both proBNP and NTproBNP circulate in human plasma and have been proposed as markers for early diagnosis of left ventricular dysfunction as well as prognostic markers of possible cardiac complications at patients with heart failure. Cellular Localization: Secreted Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
