Compromised spindle checkpoints are thought to play an important role in genetic instability that predisposes cells to malignant transformations. Human cells contain two related checkpoint protein kinases, evolved from the BUB1 gene; hBUBR1 and hBUB1. They are important components of the spindle checkpoint Both kinases monitor mitotic kinetochore-microtubule interactions. hBUBR1 is essential for normal mitotic progression as it prevents cells from entering into anaphase too early. BUBR1 has been detected in human cancers. It has been identified as a novel binding partner of BCSG1 (Breast Cancer-Specific Gene 1), which has been suggested to accelerate the progression of breast cancer by compromising the mitotic checkpoint control through inactivation of BUBR1. Cellular Localization: Nuclear Applications: Suitable for use in Immunofluorescence and Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
