Recombinant Murine C-10 is a single, non-glycosylated, polypeptide chain containing 95 amino acids and having a molecular mass of 10710 Dalton.\n\nMurine C-10 (MRP-1, Chemokine (C-C motif ligand 6, CCL6) is a member of the C-C chemokine family of proteins related to macrophage inflammatory protein-1 alpha (MIP-1 alpha). Murine C-10 is a 10.7kD protein containing 95 amino acid residues. In mice, CCL6 is expressed in cells from neutrophil and macrophage lineages, and can be greatly induced under conditions suitable for myeloid cell differentiation. It is highly expressed in bone marrow cultures that have been stimulated with the cytokine GM-CSF. Some low levels of gene expression also occur in certain cell lines of myeloid origin (e.g. the immature myeloid cell lines DA3 and 32D cl3, and the macrophage cell line P388D) that can also be greatly induced in culture with GM-CSF. However, in activated T cell lines, expression of CCL6 is greatly reduced. CCL6 can also be induced in the mouse lung by the cytokine interleukin 13. Mouse CCL6 is located on chromosome 11. The cell surface receptor for CCL6 is believed to be the chemokine receptor CCR1.\n\n\nSequence:\nThe sequence of the first five N-terminal amino acids was determined and was found to be Gly-Leu-Ile-Gln-Glu.\n\nDimers and Aggregates:\n(same/less than) 1% as determined by silver-stained SDS-PAGE gel analysis.\n\nBiological Activity:\nMurine C-10 is fully biologically active when compared to standard. The Biological activity is calculated by the chemotaxis assay using Balb/C mouse spleen MNCs. The maximum chemotactic activity was achieved at 100ng/ml.\n\nEndotoxin:\n(same/less than) 0.1ng/ug (IEU/ug) of Murine C-10.\n\nProtein Content:\nProtein quantitation was carried out by two independent methods: \n1. UV spectroscopy at 280nm. \n2. Analysis by RP-HPLC, using a calibrated solution of Murine C-10 as a Reference Standard.\n\nReconstitution:\nReconstitute the lyophilized Murine C-10 in sterile 18MO-cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.\n\nStorage and Stability:\nLyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
