Casitas b-lineage lymphoma (c-Cbl) is a 120kD cytoplasmic protein product of the c-Cbl proto-oncogene and is mainly expressed in thymus. C-Cbl is a member of ubiquitin-protein ligases (E3) family. It possesses highly conservative N-terminal phosphotyrosine binding domain that allows recognition of tyrosine-phosphorylated substrates and C3HC4 RING finger motif that allows recruitment and activation of E2 ubiquitin-conjugating enzyme. Thus, c-Cbl negatively regulates many signaling pathways that employ tyrosine-kinase receptors. Recent studies demonstrated that c-Cbl is involved in thymus development and plays a major role in whole-body homeostasis.
