c-Kit, a 145kD Type III transmembrane Tyrosine Kinase receptor, is a novel member of Ig superfamily. It consists of an extracellular domain with Ig-like repeats and a Tyrosine Kinase domain, a putative transmembrane region and a cytoplasmic domain. c-Kit functions as receptor for MGF/SCF resulting in maturation of hemopoietic and primordial germ cell precursors and melanocytes during embryonic development. The protein gets autophosphorylated and activates signaling pathways including PI3K/Akt, MAPK and JNK affecting multiple cellular functions including cell survival, proliferation, adhesion and differentiation. Mutations in c-Kit are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia and piebaldism.
