Cadherins are transmembrane glycoproteins involved in Ca++ dependent cell adhesion (1). The cadherin family, including (E)pithelial-Cadherin, (N)euronal-Cadherin, (P)lacental-Cadherin and (R)etinal-Cadherin, plays an important role in tissue morphogenesis and structure. N-cadherin (136kD) is expressed in nerve cells, developing skeletal muscle, embryonic and mature cardiac muscle cells, and pleural mesothelial cells. Studies have indicated that analysis of E-cadherin and N-cadherin expression may be useful in differentiating pleural mesotheliomas (N-cadherin positive) and lung adenocarcinomas (E-cadherin positive). Applications: Suitable for use in Immunofluorescence and Western Blot. Other applications not tested. Recommended Dilution: Immunofluorescence: 1:100 Western Blot: 1:100 detects a band at 135kD. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Additional info: Recognizes the N-terminal portion of the extracellular domain of chicken A-CAM. Species crossreactivity: human, mouse, rabbit and rat. Inhibits adherens junction formation and disrupts existing junctions of cultured lens cells. Stains a large variety of intercellular junctions formed by chicken fibroblastic and epithelial cells in culture. In adult mammals, the antibody stains predominantly heart and lens tissue and also stains a large variety of embryonic tissue. By double labelling, this antibody appears to localize vinculin and actin-rich intercellular contacts and is absent from cellular matrix attachment sites.
