The calcium-sensing receptor (CaSR) is an integral membrane protein that is a member of the G protein-coupled receptor family. It senses changes in the extracellular concentration of calcium ions. The activity of this receptor is mediated by a G-protein that activates a phosphatidylinositol-calcium second messenger system. Most cell lines do not express endogenous Ca++ sensing receptor. Studies have been done with HEK-293 or MDCK cells stabily or transiently expressed lysates. This protein is also found in native tissues. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:50-1:200 Recognizes a band at ~130kD and a large smear at the top of the gel. This protein is susceptible to aggregation, thus resulting in just seeing the high MW smear. Optimal dilutions to be determined by the researcher. Positive Control: Human Kidney Protein Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
