A unique family of cysteine proteases has been described that differs in sequence, structure and substrate specificity from any previously described protease family. This family, termed Ced-3/caspase-1, is comprised of caspase-1, caspase-2, caspase-3, caspase-4, caspase-6, caspase-7, caspase-9, and caspase-10. Ced-3 family members function as key components of the apoptotic machinery and act to destroy specific target proteins, which are critical to cellular longevity. Caspase-10 has been shown to activate caspase-3 and caspe-7 in response to apoptotic stimuli.
Applications:
Suitable for use in Immunohistochemistry. Other applications not tested.
Recommended Dilution:
Immunohistochemistry (Formalin fixed paraffin embedded): 1:100 for 10min at RT
Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10min followed by cooling at RT for 20min
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
